Although you could run either single or twin, both with almost identical results (since all aftermarket twins run in parallel), a single will be a lot cheaper. Note that you won't get a full 550rwhp out of the oem auto tranny with good reliability - that's only with the 6spd. With the auto, don't exceed about 500rwhp.

With the 6spd and 550rwhp, you'd need a clutch with more clamping force than oem (eg. SouthBend or RPS). With an auto and 500rwhp, no driveline modifications are required. You'll very likely need to run a few race fuel in every tank of gas (only when you're wanting to use the full 500rwhp+), or a meth injection kit, depending on your boost level & the size of the turbo you choose. Tuning is essential, so you'll need some kind of fuel controller, like an A'PEXi S-AFC2. You'll need wider rims & tires so you don't just sit there & spin with the additional power.

Lastly don't underestimate the value (and the cost) of doing regular & thorough maintenance on your Mkiv Supra Turbo, according to the owner's manual. Full synthetic oil, regular coolant flushes, tranny, brake fluid, diff, power steering, etc. fluid changes, alignments, etc. are all essential.

That's about all I'd suggest off the top of my head...other members please feel free to post additional suggestions...
