On a lift, with a transmission jack and two people who know exactly what they're doing, it can be done in four hours.
Alternatively, with the right tools you can do it yourself on high jackstands in about eight hours (you'll need a buddy's help during the job at two points, for about 15 minutes each time). You'll need to study the steps ahead of time, and be sure you have *everything* ready:

P.S. Don't give these instructions to 'bubba' at your local tranny shop - I can promise you he'll mess it up. A Getrag 6spd transmission is about $6K from Toyota......

X2 on not letting any shop do the change. With such a critical component, it can be messed up pretty easy. If you have the tools and know-how to do it yourself, I would suggest that.
